This category covers peptides that act on the somatotropic axis — the hypothalamic-pituitary signalling system that regulates growth hormone release.

Two mechanisms are represented. Growth hormone releasing hormone (GHRH) analogues such as CJC-1295 act at the GHRH receptor on pituitary somatotrophs. Growth hormone secretagogues such as ipamorelin act at the ghrelin receptor (GHS-R1a) through a separate pathway. Because the two act at different receptors, they are frequently studied together in the literature, which is why combination preparations are common.

A note on nomenclature that causes genuine confusion: CJC-1295 exists in two forms. The DAC (drug affinity complex) version incorporates a modification that substantially extends its half-life. The "no DAC" version — sometimes labelled mod GRF (1-29) — does not. These are different molecules with different pharmacokinetic profiles, and study results are not interchangeable between them. Check which form a paper used before applying its findings, and check which form a supplier is shipping.
